Dried Blood Spot Technique Applied in Therapeutic Drug Monitoring of Anticancer Drugs: a Review on Conversion Methods to Correlate Plasma and Dried Blood Spot Concentrations

نویسندگان

چکیده

Anticancer drugs are notoriously characterized by a low therapeutic index, the introduction of drug monitoring (TDM) in oncologic clinical practice could therefore be fundamental to improve treatment efficacy. In this context, an attractive technique overcome conventional venous sampling limits and simplify TDM application is represented dried blood spot (DBS). Despite significant progress made bioanalysis exploiting DBS, there still need tackle some challenges that limit technology: one main issues comparison concentrations obtained from DBS with those reference matrix (e.g., plasma). fact, use assays estimate plasma highly dependent on chemical-physical characteristics measured analyte, particular how these properties determine partition whole blood. present review, we introduce critical investigation DBS-to-plasma concentration conversion methods proposed last ten years applied quantitative anticancer matrix. To prove concordance between concentration, results statistical tests presence or absence trends biases were also considered.

Therapeutic Drug Monitoring by Dried Blood Spot: Progress to Date and Future Directions

This article discusses dried blood spot (DBS) sampling in therapeutic drug monitoring (TDM). The most important advantages of DBS sampling in TDM are the minimally invasive procedure of a finger prick (home sampling), the small volume (children), and the stability of the analyte. Many assays in DBS have been reported in the literature over the previous 5 years. These assays and their analytical...

Measurement and Comparison of Organic Compound Concentrations in Plasma, Whole Blood, and Dried Blood Spot Samples

The preferred sampling medium for measuring human exposures of persistent organic compounds (POPs) is blood, and relevant sample types include whole blood, plasma, and dried blood spots (DBS). Because information regarding the performance and comparability of measurements across these sample types is limited, it is difficult to compare across studies. This study evaluates the performance of POP...

Dried blood spot analysis suitable for therapeutic drug monitoring of voriconazole, fluconazole, and posaconazole.

Invasive aspergillosis and candidemia are important causes of morbidity and mortality in immunocompromised and critically ill patients. The triazoles voriconazole, fluconazole, and posaconazole are widely used for the treatment and prophylaxis of these fungal infections. Due to the variability of the pharmacokinetics of the triazoles among and within individual patients, therapeutic drug monito...

A phenytoin assay using dried blood spot samples suitable for domiciliary therapeutic drug monitoring.

A commercially available substrate-labelled fluorescent immunoassay procedure has been modified to create a simple rapid method for the determination of capillary phenytoin concentration in dried filter paper blood spots of patients on phenytoin therapy. The specimens are collected and dispatched to the laboratory by the patient, thus domiciliary monitoring of phenytoin therapy can take place. ...

Critical sample pretreatment in monitoring dried blood spot citrulline.

BACKGROUND Plasma Citrulline concentration has been correlated to functional enterocyte mass. Dried blood spot (DBS) analysis using LC-MSMS reduces sample amount needed. We optimized DBS elution to increase precision and accuracy of DBS LC-MSMS analysis. METHOD DBS control samples were eluted in varying pH (2.2-7.0) and for varying times (15-75 min) and Cit, Arg and Orn were analyzed using LC...
